- Brachial-level transverse sections of E12.5 wild-type and CNTN2 -/- (CNTN2 KO) mouse embryos were labeled with Anti-CNTN2 (TAG-1) [IPI-CNTN2.57] (Addgene #254655). No labeling is observed on CNTN2 -/- sections.

All spinal cord sections were collected from brachial level. Immunohistochemistry (IHC) on 150 μm-thick vibratome sections, 2D neural tissue explants, collagen gel COS-7 cell cultures, and Dunn chamber cultures were described previously (Suter et al., 2020), with blocking buffer used for IHC on Dunn chamber cultures, and vibratome sections altered to include 2.5% donkey serum in lieu of bovine serum albumin.
The primary antibody used for IHC was Anti-CNTN2 (TAG-1) [IPI-CNTN2.57] (Addgene #254655, 1:200).
The secondary antibody used was the Donkey anti-Rabbit IgG (H+L) Highly Cross-Adsorbed secondary antibody, Alexa Fluor™ 594 (Invitrogen, A-21207) at a 1:200 dilution of a 2 mg/mL stock for the detection of Anti-CNTN2 (TAG-1) [IPI-CNTN2.57]. Images were acquired on a Nikon Ti-E microscope or Nikon CSU-W1 SoRa Ti2-E microscope.
