Abstract
Brachial-level transverse sections of E12.5 wild-type and Punc -/- (Punc KO) embryos were labeled with Anti-IGDCC3 (PUNC) [IPI-mIGDC3.13] (Addgene #241897). No labeling is observed on Punc -/- sections.

Additional Information

All spinal cord sections were collected from brachial level. Immunohistochemistry (IHC) on 150 μm-thick vibratome sections, 2D neural tissue explants, collagen gel COS-7 cell cultures, and Dunn chamber cultures were described previously (Suter et al., 2020), with blocking buffer used for IHC on Dunn chamber cultures, and vibratome sections altered to include 2.5% donkey serum in lieu of bovine serum albumin.

Primary antibodies used for IHC were goat polyclonal antibodies against Islet1/2 (DHSB, 1:200) and Anti-IGDCC3 (PUNC) [IPI-mIGDC3.13] (Addgene, 1:200).

Secondary antibodies used were Alexa 488-conjugated donkey anti-goat (Invitrogen, A-11055) at a 1:200 dilution of a 2 mg/mL stock for the detection of Anti-goat Islet1/2, and Alexa 647-conjugated donkey anti-rabbit (Invitrogen, A-31573) at a 1:200 dilution of a 2 mg/mL stock for the detection of Anti-IGDCC3 (PUNC) [IPI-mIGDC3.13]. Images were acquired on a Nikon Ti-E microscope or Nikon CSU-W1 SoRa Ti2-E microscope.
